Effects of Environmental Factors on Growth and Betacyanin Accumulation in Red Beet Callus Culture

Abstract

|
To investigate the characteristics of callus growth and betacyanin accumulation in red beet (Beta vulgaris) callus culture, callus was induced from red beet root explant and the effects of various environmental conditions on callus growth and betalain contents were examined. Among the various sucrose concentration levels (0¡9% (w/v)) tested, callus growth was found to be maximal at 5% sucrose concentration. However, the betacyanin content of all the sucrose treated cultures was lower than in the cultures without any supplemented sucrose. Optimal callus growth was observed when cultures were raised with B5 medium, which contained 5% sucrose and KNO©ý(2,500 §·/§¤ as an inorganic nitrogen source, and light-dark conditions (16hr illumination/day). The highest betacyanin concentration was obtained when red beet callus was cultured on medium containing 5% sucrose, 2,500 §·/§¤ KN0©ý, and 10^(-4) M L-dihydroxyphenylalanine as a pigment precursor. The betaxanthin/betacyanin ratio of the red callus was higher than that of the raw red beet extract.
